The Levosulpiride is the levorotatory enantiomer of the benzamide derivative sulpiride, a specific antagonist of the dopamine D2 receptor, and also has a moderate 5-HT4 receptor agonist effect and a weak 5-HT3 receptor antagonistic effect. It is used clinically as an antiemetic, anti-gastrointestinal dysfunction drug and anti-psychotic, with high selectivity and weak antagonistic effects for dopamine D2 receptors. It has a very powerful effect on blocking apomorphine-induced vomiting in humans and dogs, alleviating apomorphine-induced tonic spasticity in rats and reducing apomorphine-induced spontaneous movement inhibition in mice. The Levosulpiride can increase dopamine turnover and increase the accumulation of dopamine metabolites, but it does not affect the activity of adenylate cyclase sensitive to dopamine.
